Celiac, Colitis and Crohns

Celiac Disease, Ulcerative Colitis and Crohn’s Disease are all autoimmune diseases, as they all result from the body developing an immune response against itself. Celiac Disease results from insensitivity to gliaden, which is a protein from wheat, barley, rye and other grains. Ulcerative Colitis and Crohn’s Disease are collectively known as Inflammatory Bowel Disease (IBD) but they affect different areas of the gastrointestinal (GI) tract. Crohn’s Disease affects mainly the end of the small intestines, but can occur anywhere between the mouth and the anus, while ulcerative colitis affects only the colon and rectum.

Since Celiac, Ulcerative Colitis are pathologies of the GI, they can all be interlinked with the gut microbiota. They may be caused by activity of the Toll-Like Receptors (TLRs), which respond to bacterial movement or byproducts. TLRs are located on the epithelial cells that line the insides of our intestines and they recognize bacterial products such as proteins, DNA or RNA. A specific receptor called TLR5 recognizes flagellin, which is a structural protein found on the bacteria flagella. When TLRs bind to their bacterial counterparts, they recognize the bacterial threat and elicit an immune response. These receptors have the power of changing downstream gene expression to generate more or less inflammatory substances such as antimicrobial agents or cytokines to battle the inflammation. In studies done on mice, when researchers removed the TLR5, the mice developed colitis. But when the defective TLR5 DNA from the genetically modified mice was transplanted to another strain of normal mice, those normalmice did not develop colitis. This study demonstrates that the development of colitis involves the gut microbiota and is independent of genetics.
